科研成果

在基于纳米技术新型药物递释系统的应用基础研究的重要进展包括:
1. 利用内源性转运机制设计靶向药物递释系统:启用了具有肿瘤、高跨血脑屏障转运能力的多个受体、转运蛋白转运途径,评价它们高效介导脑靶向的转运行为、机制;同时发现相对于大分子抗体、蛋白,分子量小的靶向功能分子的可塑性强、靶向效率高,作为靶向功能分子具有更好的药物研发和临床应用潜力。进一步,基于血脑屏障、脑胶质瘤细胞共同靶点特征,选用对两者均具有靶向性的胆碱衍生物、葡萄糖衍生物等双功能靶向分子,实现药物的脑内病灶富集的双级靶向,同时降低对正常脑、外周组织的影响。
2. 设计微环境触发的靶向治疗策略实现药物病灶靶向递释、定点释放:根据疾病的病理和生理的微环境特征,构建了组织、细胞微环境触发的靶向、控释的药物递释系统,控制药物病灶释放,为设计高效、低毒脑靶向制剂提供了理论依据。
3. 根据疾病的生理、病理特征,设计、合成、优化高分子材料,“订制”符合药物分子、疾病治疗要求的高分子材料,构建载药能力高、控制释放、生物可降解、毒副作用低的药物载体。研究重点为氨基酸为基本单位的高分子材料。
4. 采用树枝状高分子、壳聚糖等高分子载体的纳米技术提高难溶药物的口服吸收。
5. 培养的博士生1人获上海市优秀博士论文, 培养的硕士生3人获上海市优秀硕士论文。
