科研成果

在基于纳米技术的新型药物递释系统研究取得了一些成果,主要包括:
1. 主动靶向和肿瘤渗透多肽共递释的策略,提高纳米制剂在肿瘤部位的穿透。化疗是肿瘤诸多治疗环节中至关重要的一环,其成败直接影响病人的生活质量和预后,但是化疗药物难以渗透入肿瘤组织内部。为了提高药物递送系统对肿瘤组织的选择靶向性,从血管中溢出和肿瘤组织穿透的能力,本课题组提出了主动靶向和肿瘤渗透多肽共递释的策略,提高纳米制剂在肿瘤部位的穿透,围绕该策略发表系列SCI论文4篇(Biomaterials. 2014, 35:4319-4332. Mol. Pharmaceutics. 2014, 11:90?101. Biomaterials. 2013, 34:5138-5148. Biomaterials. 2013, 34: 1135-1145.)。
2. 提高细胞穿膜肽的特异性,用于脑靶向和肿瘤靶向研究。细胞穿膜肽是近年来发现的一类具有高效细胞膜穿透能力的短肽,但其缺乏细胞特异性,临床上的应用受到限制。为了解决这个问题,本课题组通过优化给药途径、优选带适合电荷的穿膜肽及采用肿瘤微环境响应的触发式穿膜肽的策略构建了三种细胞穿膜肽修饰的纳米递释系统,提高了穿膜肽的靶向性,分别用于脑靶向和肿瘤靶向研究,提高递释系统的靶向治疗效果。围绕该主题发表SCI论文4篇(Biomaterials. 2013, 34: 196-208. Bioconjug Chem. 2013, 24:419?430. Int J Pharm. 2012,436: 840-850. Biomaterials. 2011, 32: 9888-9898.)。
3. 双重靶向肿瘤新生血管和肿瘤细胞的多靶点纳米递释系统,提高肿瘤靶向治疗效果。抗新生血管疗法是化疗的主要方法,但是该方法对边缘的肿瘤细胞没有杀伤作用,甚至增加了这些细胞的侵袭和转移,所以临床疗效较为短暂,长期治疗效果更为有限。我们筛选了一系列能同时靶向肿瘤血管和肿瘤细胞的寡核苷酸和多肽,并构建了寡核苷酸和多肽介导的纳米递释系统,能起到双重靶向作用,提高治疗效果。围绕该主题发表了4篇论文(Biomaterials. 2014, 35:8215-8226. Biomaterials. 2013, 34: 5640-5650. Biomaterials, 2013, 34: 9496-9508. Biomaterials. 2011, 32: 8010-8020.)。
4. 受体介导的纳米递释系统提高治疗性多肽的入脑效率,提高阿尔兹海默症的治疗效果。本课题组设计了两种受体介导的纳米递释系统:①乳铁蛋白修饰的纳米递释系统,通过鼻腔给药的方式减少递释系统在非靶组织的摄取,并进一步通过呼吸部高表达的乳铁蛋白受体增加纳米递释系统的经鼻入脑效率。②转铁蛋白模拟肽B6修饰的纳米递释系统,静脉注射给药后通过与脑毛细血管内皮细胞上高表达的转铁蛋白受体特异性结合提高治疗性多肽的入脑效率。研究结果表明:两种纳米递释系统能高效介导中枢活性类蛋白入脑,在治疗神经退行性疾病方面具有较高应用价值。相关研究发表于药剂学的权威期刊(Biomaterials. 2013, 34: 3870-3881,Bioconjug Chem. 2013;24:997-1007)。
5. 在国际上率先开展以大豆磷脂/二油酸甘油酯为基本处方的脂质液晶纳米制剂,并系统进行了药动学和细胞转运研究,为难溶性药物的递送提供了新方法,在此基础上发表SCI论文2篇(Int J Nanomedicine. 2012, 7: 3703-3718,Int J Pharm. 2012, 424: 58-66.)。
6. 目前培养博士生2人,硕士生9人,其中已毕业4人,2人获得上海市优秀硕士学位论文。
